ABSTRACT

Objective To explore the anatomic basis for and clinical outcomes of the internal fixation which preserves the pronator quadratus (PQ) for distal radius fractures.Methods Twenty cadaveric specimens of adult upper extremity were used for this study (14 males and 6 females).The radial and ulnar lengths of PQ,the distal and proximal widths of PQ,the distances from the distal end of PQ to the articular surface of the distal radius and to the transverse line of the wrist,and the width of the bony tunnel of PQ were dissected and measured to study the anatomical features of PQ.A retrospective study was conducted of the 18 distal radius fractures which had been treated from March 2015 to March 2017 by internal fixation with T-shaped anatomic locking compression plate (LCP) with PQ preserved.They were 8 males and 10 females,with an average age of 52.7 years (range,from 28 to 65 years).According to the AO classification,there were 8 cases of type 23-A,5 ones of type 23-B and 5 ones of type 23-C1.The functional outcomes of the wrist were assessed using the Cooney scoring system at the last follow-ups.Results The PQ muscle was flat and like a right angle trapezoid with rich blood vessels.The radial and ulnar lengths of PQ were about 4.60 cm and 4.46 cm;the distal and proximal widths of PQ were about 4.41 cm and 4.48 cm;the distance from the distal end of PQ to the transverse line of the wrist was about 3.61 cm;the widths of the distal and proximal bony tunnels were about 3.08 cm and 1.91 cm.The 18 patients were followed up for 6 to 36 months (average,11.5 months).Bone union was achieved in all the patients after a mean time of 2.5 months (range,from 2 to 3 months).The mean Cooney score for the wrist function was 97.7 (range,from 95 to 100) at the last follow-up,yielding an excellent rate of 100%.Conclusions The transverse line of the distal radius fracture is located between 1/4 and 1/2 of the distal PQ.The bony tunnel of PQ is wide enough.It is feasible to preserve the distal PQ muscle in the internal fixation of distal radius fractures of types 23-A,23-B and 23-C1,because it may lead to rapid recovery of the patients and satisfactory wrist function.

ABSTRACT

Objective To explore whether augmenter of liver regeneration (ALR) can serve as early diagnostic biomarker in the patients with acute kidney injury(AKI).Methods The inpatients with possibility to AKI in the central ICU of Second Affiliated Hospital of Chongqing Medical University from October 2014 to October 2015 were recruited and assigned to the AKI group and non-AKI group according to the KDIGO guidance.Blood and urine ALR and blood creatinine were detected at 0,6,12,24,48,72 h after entering the group.Results Among all cases,40 cases(62.5%) developed to AKI.Blood ALR and urine ALR at 6h after entering the group in the AKI group began to significantly increase compared with the non-AKI group(P<0.05),the blood and urine ALR levels reached to the peak value at 12,24 h after entering the group;the blood creatinine level at 12 h after entering the group in the AKI group began to significantly increase compared with the non-AKI group(P<0.05),blood creatinine level was still slowly and progressively elevated at 72 h after entering the group.Conclusion Serum and urine ALR levels are significantly increased in the early stage of AKI,which indicates that ALR may be a new type biomarker for diagnosing AKI.

ABSTRACT

Objective To investigate the effects of D-tryptophan (D-Trp) on the formation of Streptococcus mutans (S. mutans) biofilm and the dispersal of 24 h-old biofilm, and the drug susceptibility of S. mutans against chlorhexidine (CHX) under the role of D-Trp. Methods Optical density assay was used to evaluate the growth curve of S. mutans exposed to 5.0 mmol/L D-Trp for 28 h. The non-treated group was not added with D-Trp. After treatment with 1.0, 2.5 and 5.0 mmol/L D-Trp, crystal violet staining was used to observe the changes of S. mutans biofilm formation in treatment group and non-treatment group. Crystal violet staining and confocal laser scanning microscopy (CLSM) were applied to illustrate the effects of 1.0, 2.5 and 5.0 mmol/L D-Trp on the dispersal of 24 h-old S. mutans biofilm. Resazurin sodium was used to indicate the effect of 5.0 mmol/L D- Trp on the minimum inhibitory concentration (MIC) and the minimum biofilm inhibitory concentration (MBIC) of treatment groups and negative control group. Results The growth curves of planktonic S. mutans within 28 h was consistent in treatment group and the non-treated group, both attained exponential phase after 4 h and reached stationary phase at 22 h. Notably, when compared with non-treated group, the biomass of S. mutans biofilm was increased with time from 0 to 72 h after treatment with 1.0, 2.5 and 5.0 mmol/L D-Trp. And at the same time point, the biomass was significantly less in each subgroup of treatment group than that of non-treated group (P<0.05). Crystal violet staining demonstrated that values of biomass(OD570)were less in treatment groups treated with 1.0, 2.5 and 5.0 mmol/L D-Trp than those of non-treated group (P<0.01). CLSM also showed that bacteria was adhered to the surface of media intreatment groups treated with 1.0, 2.5 and 5.0 mmol/L D-Trp. The values of biomass were lower in treatment groups than those of non-treated group (P<0.01). The MIC against S. mutans was 0.073 mg/L in both experimental group and negative control group. The values of MBIC were 0.293 mg/L and 2.344 mg/L in experimental group and negative control group, respectively. Under the action of 5.0 mmol/L D-Trp, the MBIC of S. mutans was reduced to 1/8. Conclusion Results indicate that D-Trp may inhibit the formation of S. mutans biofilm and promote the dispersal of biofilm already formed. D-Trp may further help CHX exert its bactericidal activity to S. mutans.

ABSTRACT

Objective To investigate the effects of recombinant human augmenter of liver regeneration (rhALR) on renal inflammation in acute kidney injury (AKI) induced by renal ischemia reperfusion (IR). Methods SD rats were randomly divided into sham-operated group,IR group,rhALR1 group (100 μg/kg) and rhALR2 group (200 μg/kg).Both renal pedicles of rats were identified and occluded with microvascular clamps for 60 min to induce acute kidney injury (AKI).Blood urea nitrogen and serum creatinine levels were evaluated using a Hitachi 747 automatic analyzer. For histological examination, sections were stained with HE. The activity of myeloperoxidase (MPO) was detected by spectrophotometer.Expression of TNF-α,ICAM-1,MCP-1 was determined by Western blotting. Results Blood urea nitrogen,serum creatinine levels and the injury of kidney were improved significantly in rhALR group as compared with IR group (all P< 0.05).They were improved more significantly in rhALR2 group as compared to in rhALR1 group (all P<0.05).The protein levels of TNF-α,ICAM-1,MCP-1 and the activity of MPO in kidneys from the sham-operated rats were low,and increased significantly after renal ischemia reperfusion injury (all P<0.05).After treated with rhALR,the expression of TNF-α,ICAM-1,MCP-1 and the activity of MPO were decreased significantly in kidneys as compared to those in IR group (all P<0.05),which decreased more significantly in rhALR2 group than those in rhALR1 group (all P< 0.05). Conclusions nhALR can protect kidneys from ischemia reperfusion injury in rats.The mechanism may be associated with the inhibition of renal inflammatory cells infiltration and down-regulated expressions of YNF-α,ICAM-1 and MCP-1 in the kidney.

ABSTRACT

Objective To investigate the protective effects of recombination rat augmenter of liver regeneration (rrALR) on tubular cell injury and renal dysfunction in rats with acute renal failure (ARF)induced by gentamicin. Methods One hundred fifty female Wistar rots were randomly divided into 5 groups: group A (control), group B (gentamincin 140 mg/kg+ saline 100 μg/kg), group C (gentarnincin 140 mg/kg +blank plasmid 100 μg/kg), group Dl (gentaminein 140 mg/kg+rrALR 80 μg/kg), group D2 (gentamincin 140 mg/kg +rrALR 160 μg/kg). Rats were sacrificed at day 4, 8, 12, 16 and 21 after gentamicin first injection. Blood urea nitrogen (BUN), serum creatinine (Scr) and urine N-aeetyl-β-D-glucosaminidase (UNAG) were measured. The histopathologic changes were observed by periodic aeid-Schiff (PAS) staining. Protein expression of ALR and PCNA in renal tissue was detected using immtmohistochemistry and Western blot. Results Compared with control rots, the levels of BUN, Scr and UNAG were significantly increased in ARF rats at day 4, 8, 12 and 16 (P<0.05), however, the renal dysfunction and the renal histopathologieal injury were significantly improved in ARF rats simultaneously administered with rrALR (group D) compared with ARF rats (group B and C). The protein level of ALR, the number of PCNA positive tubular ceils and the proliferation index (PI) in group D were significantly higher than those of group B and C (P<0.05). Conclusion rrALR can promote the regeneration of tubular cells in nephrotoxie ARF rats and ameliorate renal dysfunction.

Methods The cultured human renal tubular cell line (HK2 cells) was divided into several groups with various concentration of gentamicin(GM) or/and rrALR. The proliferation of HK2 cell was evaluated by 3H-TdR incorporation. The apoptosis of HK2 cells was assessed by AO/EB staining and flow cytometer using Annexin V-FITC and propidium iodide (PI) staining. ABSTRACT

Objective To develop the formulation of self-microemulsifying drug delivery system for hawthorn leaves flavonoids (HAW-SMEDDS). Methods The optimum formulations of oil phase, surfactant, and assistant surfactant for HAW-SMEDDS were screened by solubility test, compatibility test, and pseudo-ternary phase diagrams, with the time of formulating microemulsion, the consequence of visual examination, and particle size as indexes. The dissolution of HAW-SMEDDS was measured, taking the commercial tablet Yixintong Tablet as reference. Results The optimum self-microemulsifying drug delivery system was composed of Labrasol (35%), Transcutol P (10%). The particle diameter was (39.5?5.4) nm, the time of self-microemulsifying was less than 1 min. The percent of accumulated dissolution of hawthorn leaves flavonoids in SMEDDS in distilled water was up to 70% at 10 min, while that in the Yixintong Tablet was less than 50% at 60 min. Conclusion The formulation of HAW-SMEDDS preparation could meet the request of the design. It could provide the reference for the new dosage form.

AIM:To investigate the effects of recombinant rat augmenter of liver regeneration (rrALR) on apoptosis of renal tubular cells (NRK-52E cells) induced by gentamycin sulfate (GM). METHODS: The cultured NRK-52E cells were divided into four groups: normal control cells, cells with GM (GM 1.6 g/L) or GM and rrALR (15 mg/L or 25 mg/L) treatments. The apoptosis of cultured cells were assessed at 24 h, 48 h by AO/EB staining, DNA agarose gel electrophoresis analysis and flow cytometry using Annexin V-FITC and propidium iodide (PI) staining.
